Our transformants
Transformants for transactivation project.
The following table compiles lines that are believed to be homozygous.  All of these lines have the white gene as the transposition marker.  Those inserts that are identified as being on the 2nd or 3rd chromosome have been subjected to the homozygosing protocol.  Those inserts on the X chromosome were homozygosed via single pair matings.

G5 70W 4x orange X chromosome.  Southern should be rechecked.  Verified by Southern
G2 70W 1x orange X chromosome.  Southern remains to be done.  Failed verification on Southern
G2 70W 3x red X chromosome.  Verified by Southern.  Possible choice for dysgenesis.
G2 70W 4x orange X chromosome.  Verified by Southern, may have two inserts.
G2 70w 6 orange 2nd chromosome.  Failed verification on Southern.
The following table compiles additional lines that have not been subjected to the homozygosing procedure.  These lines are being maintained by inter se mating. As of 10/23/98, all of the stocks listed in the above table are also being maintained as inter se stocks.  Those lines listed in the above table are not listed in the table below.  Although some of these are designated with an X to indicate X-linkage, it would be prudent to evaluate this.

Inr A A.1.1 yellow potential new line -needs to be cut with RV/XhoI**. Homozygous
Inr A D.1.1 From dysgenisis - requires verification
Inr A D.1.2 From dysgenisis - requires verification
Inr A F.3.1 From dysgenic cross, uncharacterized
Inr A F.3.2 From dysgenic cross, linked to SM6a (curly)
Inr B (aka B.1) yellow orange 2nd chromosome. Homozygous. CPRG - moderate. Moderate X-gal staining of adult tissue (O/N). Verified by Southern.
Inr B A.1.1 light orange potential new line  Verified as transformant by Southern, but probably contains the original Inr B mutation, needs to be cut with RV/XhoI**
Inr B A.1.2 male red-orange, female orange potential new line Verified as transformant by Southern, may contain multiple inserts.  I am out-crossing to separate the inserts.

delta TA C.3 orange From dysgenic cross, linked to SM6a (curly).  Verified by Southern.
delta TA C.4 orange From dysgenic cross, linked to SM6a (curly). Needs to be cut with RV/XhoI**
70w (+25) A.1 light orange Hokuto's transformant - heterozygous
70w (+25) A.2 Hokuto's transformant - heterozygous
70w (+25) A.4 Hokuto's transformant - homozygous
70w (+25) A.7 Hokuto's transformant - homozygous.  Unusually low CPRG activity.
70w (+25) B.1 Hokuto's transformant - homozygous.
70w (+25) B.2 Hokuto's transformant - homozygous.
70w (+25) B.3 Hokuto's transformant - homozygous.
RV/XhoI **  When cut with EcoRV alone, these lines produce the desired, common length band.  However, the variable length band is very high molecular weight.  This band appears as a smear (probably because larger fragments are more suceptible to breakage and shearing).  A double digest might help reduce the size of these variable length bands and provide conclusive evidence that these lines contain new, unique inserts.

White gene transformants that have unusal variations on the hsp70 promoter.

The following table compiles what remains of some of our old transformed lines.  These lines are transformed with derivatives of our rosy vector called Car20 ZT.2.  Some of the information is based on a table I found in my lab notebook dated 7/14/95.
